The application note Analysis of axoCells Atrial and Ventricular Cardiomyocytes Electrophysiology Using CorePlate Enabled HD-MEA System presents a comparative study of Axol Bioscience’s chamber-specific cardiomyocytes using high-density multielectrode array (HD-MEA) technology.
Key highlights include:
- Electrophysiological Profiling: Atrial (ax2518) and ventricular (ax2508) cardiomyocytes were cultured on 3Brain’s CorePlate HD-MEA system. BrainWave software was used to analyze field potential duration (FPD), beat rate, and waveform morphology.
- Spontaneous Activity: By Day 7 post-thaw, both cell types formed beating monolayers. Atrial cells showed higher beat frequency, while ventricular cells exhibited greater T-peak amplitude, indicating distinct repolarization dynamics.
- Isoproterenol Response: Atrial cardiomyocytes responded to 10 µM isoproterenol with increased beat rate, shortened QT interval, and elevated R–S and T-peak amplitudes, consistent with β-adrenergic stimulation.
- HD-MEA System: The CorePlate 1W 38/60 features 4,096 electrodes in a 3.8 × 3.8 mm² area, enabling high-resolution mapping of cardiac activity and pacing capabilities.
- Assay Workflow: A 7-day thaw-to-data protocol was used, with cells plated on fibronectin-coated CorePlates and maintained in Cardiomyocyte Maintenance Medium.
This platform supports high-throughput, chamber-specific cardiac electrophysiology for drug screening and arrhythmia research.
